Structural domains comprising this superfamily share the structure of the C-terminus of NifE, an essential player in nitrogenase MoFe cofactor biosynthesis. The iron molybdenum cofactor (FeMo-co) is considered to be the site of dinitrogen reduction and it has been proposed that NifNE, which arises via interaction of NifN and NifE proteins, serves as a scaffold upon which FeMo-co is built.
