Structural domains comprising this superfamily share the structure of the C-terminal domain of a group of neutral/alkaline ceramidases found in both bacteria and eukaryotes PMID:10753931,PMID:10781606,PMID:10593963 hydrolyzes ceramide to generate sphingosine and fatty acid. The enzyme plays a regulatory role in a variety of physiological events in eukaryotes and also functions as an exotoxin in particular bacteria PMID:19088069.

Ceramidases play a key role in sphingolipid metabolism by converting the apoptosis-associated lipid ceramide to Sph, a precursor for the proliferative factor S1P. Three families of CDases (acid, neutral, and alkaline) have been identified that are distinguished by their pH optima, subcellular localization, primary structure, mechanism, and function PMID:26190575. This entry represents the C-terminal domain of both neutral and alkalyne non-lysosomal ceramidases. indispensable for the correct folding, localization, and enzyme activity; however, the molecular basis is poorly understood PMID:15123644,PMID:19088069.
