Functional Families

Overview of the Structural Clusters (SC) and Functional Families within this CATH Superfamily. Clusters with a representative structure are represented by a filled circle.

Superfamily: Spidroin domain, C-terminal domain

Spider silk fibres are made from spidroin proteins, notably aciniform spidroin 1 (AcSp1), minor ampullate spidroin (MiSp), major ampullate spidroin (MaSp) and tubuliform spidroin 1 (TuSp1). Dragline silk is composed of two proteins, major ampullate spidroin 1 (MaSp1) and major ampullate spidroin 2 (MaSp2) PMID:16283740. MaSp1 contains five alpha-helices PMID:17096540.

Spidroins contain repetitive units capped between non-repetitive N-terminal (NTD) and C-terminal (CTD) domains, whose composition varies among species and thus determines the variety of mechanical properties of spider silks. The CTDs are conserved among spider silk proteins. This superfamily comprises spidroin-like CTDs largely present in invertebrates, particularly arachnids.
