FunFam 11: Homogentisate phytyltransferase 1

There are 3 EC terms in this cluster

Please note: EC annotations are assigned to the full protein sequence rather than individual protein domains. Since a given protein can contain multiple domains, it is possible that some of the annotations below come from additional domains that occur in the same protein, but have been classified elsewhere in CATH.

Note: The search results have been sorted with the annotations that are found most frequently at the top of the list. The results can be filtered by typing text into the search box at the top of the table.

EC Term Annotations Evidence
Homogentisate phytyltransferase. [EC: 2.5.1.115]
Phytyl diphosphate + homogentisate = diphosphate + 2-methyl-6- phytylbenzene-1,4-diol + CO(2).
  • Involved in the biosynthesis of the vitamin E tocopherols.
  • While the enzyme from the cyanobacterium Synechocystis PCC 6803 has an appreciable activity with geranylgeranyl diphosphate (EC 2.5.1.116), the enzyme from the plant Arabidopsis thaliana has only a low activity with that substrate.
  • Formerly EC 2.5.1.n8.

Coumarin 8-geranyltransferase. [EC: 2.5.1.138]
(1) Geranyl diphosphate + umbelliferone = diphosphate + 8-geranylumbelliferone. (2) Geranyl diphosphate + esculetin = diphosphate + 8-geranylesculetin.
  • The enzyme, characterized from the plant Citrus limon, is specific for geranyl diphosphate as a prenyl donor.
  • It also has low activity with the coumarins 5,7-dihydroxycoumarin and 5-methoxy-7-hydroxycoumarin.

Umbelliferone 6-dimethylallyltransferase. [EC: 2.5.1.139]
Dimethylallyl diphosphate + umbelliferone = diphosphate + demethylsuberosin.
  • The enzyme from parsley (Petroselinum crispum) is specific for umbelliferone and dimethylallyl diphosphate.
  • A minor product is osthenol, which is produced by transfer of the dimethylallyl group to C-8 of umbelliferone.
