In eukaryotic replication licensing, Cdt1 plays a key role by recruiting the MCM2-7 complex onto the origin of chromosome. This superfamily entry represents the C-terminal domain of Cdt1 (mCdt1C characterised in mice). It is the most conserved region in Cdt1, is essential for licensing and directly interacts with the MCM2-7 complex PMID:19722278. The C-terminal region contains a WHD (winged helix domain), a typical DNA recognition motif PMID:20335175. The winged-helix fold is conserved throughout several other licensing factors, suggesting that they evolved from a common ancestor. In mouse Cdt1 (mCdt1C) the main role of the WH motif is to interact with MCM2-7 rather than with the DNA, as other winged-helix domains.
